Yes you can drive with a UK licence how would people be able to hire Spanish cars otherwise.

As the car is my brother in law who lives in Spain and before UK came out of the eu ,I drove the car when I was in Spain and insured on his policy,

Would this still be the same as the UK are out of the eu.

As a tourist, your licence is valid.  As long as his insurance doesn’t require named drivers, you’ll be fine; e.g. mine covers the car for any driver over the age of 23.  That’s pretty standard so I guess you’d be ok

To be sure when we let a friend drive ours we gave them a letter to say they had our permission.
